This book demonstrates that the PPAR- ¿ agonist troglitazone (TRO) exhibited anti-inflammatory and anti-fibrotic properties in a murine model of BLEO-induced lung fibrosis and in vitro on human lung fibroblasts. As a preventive treatment for BLEO-induced lung fibrosis, TRO has demonstrated comparable abilities to reduce lung inflammatory cytokine levels, pulmonary recruitment of inflammatory cells and collagen deposition, loss of body weight and mortality rate. TRO demonstrated anti-fibrotic effects in reducing lung collagen/HYP levels during therapeutic treatment, whereas DEX was incapable of reducing these effects. TRO also exhibited anti-fibrotic properties in vitro by inhibiting TGF ¿-¿induced -SMA protein expression and collagen secretion in lung fibroblast cultures. These results support the potential use of PPAR- ¿ agonists as therapeutic agents in the treatment of lung conditions characterised by inflammation and fibrosis.
